If you didn't notice already, there aren't any times listed on the Game Dates this year. I'm not going to take the time to go through and look at when each team starts their games this year, so you can change your lineups up until a particular game starts, with a hard deadline of 6:00 PM (12:00 on Saturdays and Sundays for the most part, there are probably exceptions).
Please note that backups come in to play here too. You can't take a guy out or put in a guy in any position (including backups) if their game has started.
I don't anticipate there will be many issues with this. In most cases, I would suggest putting in lineups early anyway.
